Bacillus subtilis YtvA expression cassette
The ytvA cassette is designed to produce the protein YtvA, which is a light-activated protein. The gene of interest (BBa_K3889026) is placed downstream to the constitutive promoter of Bacillus subtilis P43 (BBa_K143013) and the ribosome binding site of Bacillus subtilis (BBa_K1351031). ytvA is placed upstream to the porcine teschovirus-1 P2A Peptide Linker (BBa_K3889069) and mCherry (BBa_J06504). The mCherry gene has been placed to act as a reporter for the functioning of the cassette. This will give continuous red fluorescence which can be measured with the help of a plate reader. At the end of this cassette, a double terminator (BBa_B0015) has been put to terminate the transcription.
This cassette has been designed as a part of a visible light kill switch genetic circuit in Bacillus subtilis bacteria. This working of the cassette can be checked by the mCherry RFP that is placed downstream of the YtvA producing gene.
