The purpose of this part is to encode a fast degrading GFP. Wildtype GFP typically have a half-life that may last for several bacterial generations. However, the addition of an AANDENYALAA tag to the C-terminal of the wildtype GFP will decrease this halflife to 40 minutes (Andersen et al, 1998). By changing the last three residues of this tag, various fast-degrading GFP were created. GFP[LVA] has a halflife of 40 minutes.

{˂GFP-LVA˃} Fast-Degrading GFP

The purpose of this part is to encode a fast degrading GFP. Wildtype GFP typically have a half-life that may last for several bacterial generations. However, the addition of an AANDENYALAA tag to the C-terminal of the wildtype GFP will decrease this halflife to 40 minutes (Andersen et al, 1998). By changing the last three residues of this tag, various fast-degrading GFP were created. GFP[LVA] has a halflife of 40 minutes.

This part follows the BglBricks standard. It is flanked by BamHI and BglII sites instead of XbaI and SpeI. More information about the BglBricks Standard is available at:
