This part corresponds with an Ribosome Binding SIte sequence specifically designed for the cyanobacteria Synechocystis PCC6803.  This RBS corresponds with a SD consensus sequence for PCC 6803 allowing up to 5-fold higher expression than the typical E. coli AGGAGG consensus sequence.

The part has been assembled in accordance with the design guidelines of Marburg Collection https://2019.igem.org/Team:Marburg/Part_Collection. Despite that the part sequence does not include the resulting MoClo Overhangs after level 1 assembly. Then the level 0 part includes the overhangs TACTAGAG  in the 5’ end of the sequence, and the bases TAATCAATG in the 3’ end of the sequence.

The RBS was designed via an analysis of the RBS sequences of all coding genes longer than  100 amino acids in Synechocystis PCC 6803. It was found that only 26% of the genes contained the core SD sequence, while in E. coli it was 57% (Ma et al., 2002). Based on that the consensu RBS sequence TAGTGGAGGT which is complementary to the anti-SD sequence AUCACCUCCUUU of Synechocystis PCC 6803 has been designed.
