RiboJ comprises of the sTRSV-ribozyme with anadditional 23-nt hairpin immediately downstream to help expose the RBS. sTRSV autocatalytically cleaved the mRNA at a defined residue, which had the effect of cleaving extraneous RNA leaders that arose from transcription from promoters with different start sites.This can insulate the output from the influence of the genetic context, meaning that the output promoter depended only on the activity of the input promoter parts, and not the sequence at the part junction.
