Usage and Biology
Glyphosate can be metabolized into formaldehyde in E.coli. Formaldehyde promoter provides an inducible scheme for downstream genes and proteins. The inverter(BBa_Q04510) plays the role of gene inversion which reverses expression pattern when existing. MazF(BBa_K1720002) was designed after formaldehyde promoter and inverter, made sense of our kill switch system in degradation: formaldehyde promoter can be activated after formaldehyde induction and MazF expression is blocked and reversed by inverter. In contrast, without formaldehyde, MazF can express under the pR promoter inside inverter and cause cell death by protein toxicity.
