The PhoBR genetic circuit involves the usage of the PhoBR genes alongside promoter pPhoA, and repressible protein TetR. This system is coupled with the p-tet SCRIBE system (BBa_K3424023).The PhoBR genes will be activated by the Pho regulon in E.coli when these organisms lack sufficient phosphate.As PhoB gets activated, it becomes a positive regulator for itself, PhoR, and promoter pPhoA. When E.coli have sufficient phosphate, they induce the inactivation of this specific phosphate transport system. By having pPhoA coupled with TetR, we have designed a system that uses a genetic inverter so that it can detect phosphate levels beyond the regular threshold at which bacteria usually stop the gene activation of this system, allowing us to quantify this output through permanent mutations using our p-tet-SCRIBE composite(BBa_K3424023).
