Antitoxin part of a toxin-antitoxin system of B. subtilis
YqcF is a small protein acting as an antitoxin in a type II toxin-antitoxin (TA) system described in Bacillus subtilis. This protein is capable of binding to its counterpart YpcG, thereby preventing its DNase activity in the cell. Most likely, this interaction occurs through binding to the LXG domain of YpcG (Holberger et al., 2012). However, it has been shown that it doesn’t exhibit antitoxic effects on other toxins that contain the same LXG domain (REF). For our iGEM project we intend on expressing this antitoxin under the control of a solanine dependent promoter with the purpose of rescuing cells that are located in the proximity of the potato root exudate. Solanine is a main and specific component of potato root exudates and, by putting the production of YpcF under the control of solanine dependent promoter, we can only allow its production in the potato environment. The cells will constitutively express the toxin part of the system (YqcG) but their survival will be dependent on the presence of solanine. By this, we hope to implement a kill switch system that allows the B. mycoides to survive only in the designated area.
