It's noteworthy that, in our Protein Crosslinking design, we intended to use double CBM in order to anchor the CBM and SpyCatcher003 firmly. Moreover, CBM would be expressed in both P. Pastoris and E. coli, respectively. Therefore, codon optimization must be applied in order to avoid homologous recombination and fit the chassis organism. As a result, we synthesized 4 versions of genes coding the same CBM sequence based on the 2 versions of codon optimization for P. Pastoris and 2 versions for the codon optimization of E. coli. Names for those 4 versions of CDS are documented from CBM optimized V1 to CBM optimized V4, in which the CBM optimized V1 and V2 are for P. Pastoris, and CBM optimized V3-4 are for E. coli.

CBM optimized V3

The cellulose binding module (CBM) help connect cellulose and Spycatcher003.

Pastoris, and CBM optimized V3-4 are for E. coli.
