Contribution

Compared to the existing part BBa_I732005 which is a full-length lacZ, we insert hERα and intein into the lacZ to produce a new part BBa_K3525000, giving it a new function of detecting the presence of estrogenic chemicals in environmental water.

We obtained a chimeric protein composed of S. cerevisiae VMA intein, the esterogen binding domain of hERα and lacZ reporter protein. This protein may provide a complete and independent biosensor for estrogen molecular. In theory, estrogen dependent self splicing and production of active lacZ protein can be realized by this chimeric protein. The related domain parts and design principles may also be used for sensor proteins of other molecules.
