The promoter of patzD can sense the presence of atzR protein and express EGFP protein, and then emit green fluorescence.
Contribution
BBa_K3526005 is a bio-sensor that is regulated by the transcription factor PatzD. This part should be used with the part BBa_K3526004, which can consecutively express a transcription factor AtzR. Under the presence of cyanuric acid (CYA), the AtzR can bind CYA and regulate the promoter PatzD. As a result, the GFP is expressed and emit green fluorescence.
Engineering Success
To test the function of part BBa_K3526005, we co-transform the part BBa_K3526005 and part BBa_K3526004 into Pseudomonas putida KT2440, choose 0μM, 50μM, and 100μM cyanuric acid (CYA) to induce the system. It is found that the reaction has a specificity GFP signal when the concentration of CYA is 100μM.
