GFPnano-FKBP-FKBP
This part is designed to generate GFP degradation with improved rapamycin responsiveness.
Usage and Biology
FKBP is a kind of protein which can bind with FRB induced by rapamycin. GFPnano is the nanobody of GFP, a green fluorescent protein, which can be observed easily. This part could target GFP and bind TRIM21-FRB thought the FRB-FKBP interaction and lead to the degradation induced by ubiquitin-proteasome. Here is the mechanism of the recombined TRIM21-FRB:
1. The GFPnano tagged with FRB combines with targeted protein.
2. TRIM21-DocS connect Coh2-GFPnano-target through the rapamycin induced DocS-Coh2 interaction.
3. The targeted protein is degraded by ubiquitin-proteasome system recruited by TRIM21.
