Coh2 is a kind of protein which bind with DocS spontaneously. GFPnano is the nanobody of GFP, a green fluorescent protein, which can be observed easily. This part can target the protein and connect with TRIM21-DocS and lead to the degradation in a Trim-Away alike method. Here is the mechanism of the recombined TRIM21-DocS:

GFPnano-Coh2

To make sure that GFPnano-GFP can connect with TRIM21-DocS, we designed the part which bind Coh2 and GFPnano.


Usage and Biology

Coh2 is a kind of protein which bind with DocS spontaneously. GFPnano is the nanobody of GFP, a green fluorescent protein, which can be observed easily. This part can target the protein and connect with TRIM21-DocS and lead to the degradation in a Trim-Away alike method. Here is the mechanism of the recombined TRIM21-DocS:

1. The GFPnano tagged with Coh2 combines with targeted protein.

2. TRIM21-DocS connect Coh2-GFPnano-target through the DocS-Coh2 interaction.

3. The targeted protein is degraded by ubiquitin-proteasome system recruited by TRIM21.
