Design Notes

The sequence from BBa_I15009 was taken and subject to the IDT codon optimisation tool. During optimisation, no illegal restriction sites were created allowing for the part to be used at RFC[10] and RFC[1000] standard.

The pcyA gene is found in Synechocystis sp. PCC 6803 and has been previously categorised in BioBrick form by J.J. Tabor (BBa_I15009). In order to maximise expression of phycocyanobilin:ferredoxin oxidoreductase in the chosen E.coli chassis, the St Andrews iGEM team of 2020 carried out codon optimisation using the IDT codon optimisation tool.
