cMyc-Tag (TypeIIS adapted)
C-myc tag was added at the C-terminal site of ompT protein (part BBa_K3122007)to allow subsequent detection by Western Blot, since no antibody has been developed against ompT yet. We designed a new scar compatible with the assembly for this purpose, as MoClo did not suggest anyone for C-terminal tags. Although cloning of this part has not been accomplished by our team because of the lack of ompT, we think this contribution will be rather useful for future teams aiming to perform MoClo assemblies.
Design
The new scar added to iGEM type IIS standard is the following:
Left tag scar: 5' AGCC 3' Right tag scar: 5' GCTT 3'
This scars allows positioning the tag at the C terminal site of the CDS protein.
