Description

TeLuc is a triple mutant of NanoLuc. It bypasses the absorption of Chlorophyll A, making it the more suitable reporter for phototrophic organism.

This part is contained in a collection of reporters for Synechococcus elongatus UTEX2973 from iGEM Marburg 2019. They are specifically chosen for cyanobacteria as they show a different absorption and excitation behaviour compared to other bacterial cells due to their photosystems. Hence not every fluorescence based reporter can be recommended for the use in cyanobacteria. This set should be seen as a recommendation of reporters based on experimental dataand literature. They are either fluorescence reporters that bypass the wavelength at which cyanobacteria absorb and emit light or luminescent reporters that show a greater signal strength over background signal.

Compability

These parts are compatible with the RCF [1000] standard and can be used in any part collection that uses the PhytoBrick standard of overhangs. For more information we recommend to head over to Design of the Marburg Collection.
