Tac Promoter (including two operator genes(lacO))
Tac promoter is a hybrid of two operons: the trp and lac operons. Tac promoter is inducible by IPTG and commonly used in Corynebacterium glutamicum for overproduction of proteins. Corynebacterium glutamicum constitutively produces the LacI protein, a strong Ptac promoter repressor which can bind with lac operator. In our experiment, we add another operator gene to tac promoter in order to regulate the expression of the racE gene to produce γ-PGA with different D/L-glutamate monomer ratios.
