Type I CRISPR/Cas system surveillance complex Cascade ABCDE

This is a part consisting of all cofactors necessary for the functionality of the CRISPR/Cas3 system. It works if utilised with BBa_K3096001 and the respective CRISPR arrays targeting the to-be-degraded sequences.

Usage & Biology

In the type I CISPR/Cas system the crRNA guides the surveillance complex Cascade ABCDE to complementary DNA sequence. The DNA is unwound, generating the so-called R-loop. The Cas3 helicase-nuclease protein attaches to this R-loop and unidirectionally degrades the DNA. Hereby, the large Cas3/Cascade complex pulls the substrate DNA towards itself, introducing single strand breaks.
