LEE5_variant-1_ΔTir
LEE (locus of enterocyte effacement) is a genomic island that encodes a secretion system in enteropathogenic E. coli. This BioBrick is a small part of the island containing the promoter region of the 5th operon.
Compared to LEE5_native (BBa_K2688010), this variant has a point mutation of indeterminate significance. This does not preclude heterogeneous expression. This variant also has a deletion that removes the tir fragment. (The beginning of an ORF is present in the native sequence : it is removed for improved expression and to avoid clash of reading frame)
This region has been shown to be recognized by two nucleoid-associated proteins, H-NS and Ler (‘LEE encoded regulator’). The regulation by these proteins can generate some heterogeneity of transgene expression within E. coli populations as recently shown by Leh and collaborators (Leh et al, mBIO, 2017).
