Applications of BBa_K2515003
We used this part instead of BBa_K145201 for fusions with GFR generator (BBa_K081012) and RFP generator (BBa_K081014). It functioned as expected obtaining lower basal level of expression in the high copy number vector pSB1C3.
We believe that the major reason for the improvement is the removal of the LVA-like tag from TetR and the inverted orientation is not so critical but this hypothesis needs further experiments to be proven.
We also cloned this part together with a reversed transcriptional terminator (Part:BBa_B0025). We did not observe a functional improvement.
