Temperature-sensitive repressible system
In this construct, the temperature sensitive TlpA36 protein is constitutively produced, and depending on the temperature it will either form dimers capable of repressing the pTlpA36 promoter (in temperatures below 36 degrees) or stay in monomeric form which is unable to repress the promoter.
As a proof of concept that the temperature system is fully functional, we had constructed and characterised an ITPG-inducible temperature system and coupled it to a downstream GFP reporter.
