Lycopene cyclase downstream in PCC7942

In order to present Lycopene in Cyanobacterium Synechococcus sp Strain PCC7942, we use the strategy:“Gene Knock Out”. This part provides the sequence of the downstream of Lycopene cyclase in Cyanobacterium Synechococcus sp Strain PCC7942 (we call it ” Lycopene-DS”) Lycopene-DS is one of the parts to construct the plasmid executing "Gene Knock Out".

When the plasmid has been constructed completely, it contains ” Lycopene-US + AmpR + Lycopene-DS”.(As you can see, Part BBa_K2350014) There is a probability to occur Homologous Recombination after transferring this plasmid into PCC7942. The Lycopene cyclase gene (CrtL) will be replaced by Ampicillin Resistance gene(AmpR) in Homologous Recombination. And then, the “Gene Knock Out” is completed.
