In order to characterize the transfer function of a promoter with and without RiboJ, it is necessary to induce the activity of that promoter. plLacO-1 and pTac are both repressed by the lac repressor, which in turn is repressed by IPTG— hence IPTG will induce the activity of plLacO-1 and pTac. This part codes for constitutive expression of the Lac repressor protein. The part is flanked by the USN 2 and 3 sequences as per the WM UNS standard.
