<br>We select the strong constitutive promoter J23119 and strong RBS B0034 to express inhibitor of pTet, tetR. And we ligate induced promoter pTet following the inhibitor. In order to decrease impact between constitutive promoter and induced promoter, we reserve the constitutive promoter’s direction and the transcription of it will not impact the downstream killer. Then killer, ''mazF'', which locates at downstream of induced promoter will be regulated by concentration of inhibitor connecting plasmid copy numbers in a suitable range. Then this device can kill the cell with little plasmids and maintain a high expression level of function gene in industrial production.

<Description>
This is one of our application circuits.


This circuit contains two devices: inhibitor device and killer device.
Inhibitor device: We select strong constitutive promoter J23119 and strong RBS B0034 to express inhibitor gene tetR.
Killer device: Toxin protein, MazF, can cleavage ACA sequence of free mRNA specifically. It cause bacteria can’t translate successfully and dead finally. So we constructed it with induced promoter pTet and medium RBS B0032 to control the expression level of toxin protein.
We reversed the direction of inhibitor device, in order to reduce the influence between two devices.
