Synthetic Enhancer Project: 3X TetO Binding Cassette(52S)
52S binding cassette has three TetO binding sites. Based on the amount of aTc in the system, which binds and inactivates TetR repressor, the amount of Tetr bound to these two TetO varies and thus flexibility of the DNA is also changing and ultimately the looping between the enhancer and promoter and output levels. These three TetO sites on 52S allows for four discrete states: unrepressed, two intermediate steps, completely repressed. This part is from a plasmid gifted by Amit et. al 2011.
