This composite part consists of LacI under constitutive promoter, and toxin gene of toxin N-acetyltransferase GCN5 (In our project, ID: 1204) under pTac prompter.

Directly upstream of coding region of toxin gene is RiboJ, an insulator buffers gene expression from its cellular context. RIboJ is a ribozyme which effetely cleaves 5'-UTR of mRNAs.
