This composite BioBrick was used as a visual reporter gene. The brief half life enables this part to be used as a method to quantify protein production in a much more precise manner than a regular RFP that aggregates in the cell. A microplate reader was used to report objective data about the production of this protein. As a result of the constitutive promoter and short temporal lifespan, this part was used as an effective assay of plasmid maintenance where the transcription and translation of this RFP reflected the maintenance of a plasmid that contained this part. Alongside of this visual reporter, an additional plating test was used to verify the presence of the plasmid of interest in the E. coli samples. The results of our plating tests demonstrate a strong correlation between RFP production and number of cells that contain the PSB1C3 plasmid. This additional assay provides more evidence to verify the effectiveness of this protein over many generations as a reporter of protein production.
