This tfa circuit was synthesised removing the illegal restriction sites to make the sequence BioBrick-compatible. The sequence was cloned into a pSB1C3 and the success of the cloning procedure was confirmed by restriction with EcoRI and PstI restriction enzymes and followed by DNA agarose electrophoresis (Figure 1).
