This reporter carries a mutant of RBS BBa_B0034. This mutant carries a single base pair substitution on the second base pair. The twelfth base pair A is substituted to T. The construct consist of the pLac promoter BBa_R0011, mutant RBS, eGFP BBa_E0040 and double terminator BBa_B0015. The purpose of this construct is to characterize the mutant RBS in Shewanella oneidensis MR1 strain.

GFP Reporter with Mutant RBS Opt

This reporter carries a mutant of RBS BBa_B0034. This mutant RBS carries a total of 7 base substitutions. This RBS mutants is mutated based on the results of our BBa_B034 mutant library. Substitutions of basepairs with the highest GFP expression is carried out. The construct consist of the pLac promoter BBa_R0011, mutant RBS derived from BBa_B0034, eGFP BBa_E0040 and double terminator BBa_B0015. The purpose of this construct is to characterize the mutant RBS in Shewanella oneidensis MR1 strain.

As we can see mutations on basepair 5, 6, 7, 8 and 9 brings to a lower GFP expression. Hence, no substitutions are made on these basepairs.
