GFP Reporter with Mutant RBS 2AG

This reporter carries a mutant of RBS BBa_B0034. This mutant carries a single base pair substitution on the second base pair. The base pair A is substituted to G. The construct consist of the pLac promoter BBa_R0011, mutant RBS derive from BBa_B0034 , GFP BBa_E0040 and double terminator BBa_B0015.

The purpose of this construct is to characterize the mutant RBS in Shewanella oneidensis MR1 strain.

This reporter can also be derived from the existing construct, BBa_J69511.

The characterization is shown below,

It is shown to have a fluorescence reading higher than the wild type RBS reporter, BBa_J69511.
