Lactate sensing system

In the system, promoter P170 has been creatively constructed into E.coli to control the certain concentration of lactate. Besides, the activation of P170 cannot achieve without the help of rcfB and in order to measure the final result as convenient as possible, we also introduce a RFP reporter gene into the plasmid. Consequently, when acidification reaches a threshold, activated trans-acting protein RcfB ( BBa_K1696001) will bind to ACiD-box and lead to P170 activation. Thus, we can see the red fluorescence without any pretreatment. The characterization of this part was done using a P170-promoter + RFP construct.
