We add RFP reporter gene after P170 promoter. First, we want to test whether the inducible promoter is leaky or not. Then we use RFP gene to characterize the lactate-sensing system's performance in E.coli.

P170 is a acid-induced promoter, however, its trans-acting protein, RcfB, is involved in the basal activity leading to the leakage[1]. To test whether the inducible promoter is leaky or not and characterize the lactate-sensing system's performance in E.coli, we add RFP reporter gene after P170 promoter. Red fluorescent protein (rfp), isolated from a sea anemone relative (Discosoma sp.), is an ideal reporter protein for living color. Because its fluorescence can be easily detected without any pretreatment[2].
