rpoN: coding sigma 54 factor
This part codes sigma 54 factor of E.coli and its RBS. We use this part to make nutritional switch which is activated under poor availability of combined nitrogen. The complex of sigma 54 factor and the other transcription factor activates sigma 54 promoter.
Usage and Biology
Sigma-54 plays a role in quorum sensing of Vibrio harveyi, which uses AutoInducer-2(AI-2) as the quorum sensing molecule. At conditions of low cell density(when AI-2 is low), LuxQ with the periplasmic protein LuxP transfer phosphate to LuxO. LuxOP in conjunction with Sigma54 indirectly represses expression of the genes encoding luciferase (lux), so no light is produced under this condition. At low cell density, LuxO∼P, together with Sigma54, activates the expression of the genes encoding four small regulatory RNAs (sRNAs) called Qrr1-4(Quorum regulatory RNA's).
