Contains an IPTG-sensitive promoter (BBa_J04500), with RBS.3 (BBa_00032), and reporter GFP (E0040) downstream with an additional RBS and the EmrR mutator gene. Using a fluctuation analysis mutation rate experiment, XL1Blue bacteria transformed with this construct yielded a mutation rate of 3.14E-09 mutations per cell per culture although mutation rate is dependent on growth conditions and can vary depending on experimental setup. This plasmid appears to incur a growth penalty in transformants.
