R0082-HfiA the HfiA protein express passway which can be regulated by light
Introduction
HfiA is an inhibitor in the holdfast's polysaccharide synthesis pathway. It can inhibit one of the Hfs protein family-HfsJ. So it can block up the synthesis of polysaccharide. HfiA, It is regulated downstream of lovK-lovR and the relationship is as following: [Fig 1]
These results support a model in which HfiA inhibits holdfast development via direct interaction with an enzyme required for holdfast bio-synthesis.
These results support a model in which HfiA inhibits holdfast development via direct interaction with an enzyme required for holdfast bio-synthesis.
Circuit
In order to let HfiA be regulated by the upstream light sensor system, we add the same promoter as the CI coding domain to its gene. Thus the motion control protein can work with fluorescent proteins in phase. The circuit is as following.[Fig 2]
Get this part
To get PompC-HfiA, we first PCR the HfiA from genome of C.Crescentus. Then we standardize it and add the promoter to this part. We sequenced it and the result is exactly right.
