The Spinach2 sequence comes from [http://www.nature.com/nmeth/journal/v10/n12/full/nmeth.2701.html Strack ''et al.'']. This BioBrick is a modified version of Spinach2 that has been modified to remove an internal SpeI site. Spinach2.1 has been confirmed to function equally well as Spinach2. See [http://2014.igem.org/Team:DTU-Denmark/Achievements/Parts this page] for more details.

Design Notes

The sequence was synthesised, and amplified using High-Fidelity PCR. The PCR product was cut and ligated with pSB1C3.

The fusion with tRNA was designed as described in [http://www.ncbi.nlm.nih.gov/pubmed/17558412 Ponchon & Dardel]. The Spinach2 sequence comes from [http://www.nature.com/nmeth/journal/v10/n12/full/nmeth.2701.html Strack et al.]. This BioBrick is a modified version of Spinach2 that has been modified to remove an internal SpeI site. Spinach2.1 has been confirmed to function equally well as Spinach2. See [http://2014.igem.org/Team:DTU-Denmark/Achievements/Parts our wiki] for more details.
