Many signalling cellular processes occur using a phosphorylation mechanism. In order to assemble a transmembranal pathway, that frees a phosphore, with a genetic network within the cell, we use the promoter pOmpR (R0082). In E. coli, the osmosensing machinery using the EnvZ membrane protein follows this mechanism. That promoter is regulated positively (activated) by the transcription factor OmpR-P. We put a well known protein downstream: GFP (E0040) with a RBS (B0034) and a Terminator (B0015). This composite part will be very useful to characterize many cellular biosensors.

Promoter activated by OmpR-P with the reporter GFP

We use the promoter pOmpR BBa_R0082 to interface with sensors that use a synthetic two-component signal transduction pathway based on an EnvZ domain (and a EnvZ- strain such as BBa_V1012). That promoter is regulated positively (activated) by the transcription factor OmpR-P (phosphorylated).
