sgRNA (1): CRISPR Silencing of YFP

Single guide RNA1 is an RNA oligo that associates with dCAS9 (BBa_K1323002) and guides the complex to the coding strand of the YFP (BBa_K1323010) at position 43 - 62. sgRNA is a fusion of tracrRNA (trans-activation crRNA) and crRNA (CRISPR RNA) - the 20bp sequence is complementary to the target sequence and is linked to a hairpin loop that is similar to tracrRNA. When the sgRNA-dCas9 complex binds to the target sequence, it is expected to act as a programmable repressor and silence expression of the target gene.
