Silencing sRNA (10 bases long) for USP45 (Based on Spot42)
This is a mutagenesis of the antisense site of the small RNA Spot42.
It contains the J23101 promoter.
Biology
The sRNA has its base from the gene spot42. It consists of an antisense region that recognizes a specific sequence and another region that recruits the protein Hfq. The Hfq protein blocks and prevents the targeted mRNA from binding to the ribosome. This will stop the translation of the USP45 protein complex and no protein should be produced. We have redesigned the antisense region so it will specifically recognize our USP45. The antisense region is ten nucleotides long. The antisense region will make it bind to our USP45 and recruit the Hfq protein to stick to that region. USP45 is a secretion tag that we coupled to colicins Fy. But in theory it should work with anything you couple to USP45.
