+ | This is a mutagenesis of the antisense site of the small RNA [https://parts.igem.org/Part:BBa_K864440 spot42]. It contains the J23101 promoter [https://parts.igem.org/Part:BBa_J23101 J23101 promoter].

+ | The sRNA has its base from the gene spot42. It consists of an antisense region that recognizes a specific RNA sequence and another region that recruits the protein Hfq. The Hfq protein blocks and prevents mRNA from binding to the ribosome. This will stop the translation of the USP45 protein complex and no protein product should be produced.
+ | We have redesigned the antisense region so it will specifically recognize our USP45. The antisense region is ten nucleotides long. The antisense region will make it bind to our USP45 and recruit the Hfq protein to stick on that region. USP45 is a secretion tag that we coupled with colicins Fy (https://parts.igem.org/Part:BBa_K1381018). But in theory it should work with whatever you couple to USP45.
