Design Notes

Primers with RFC25 prefix and suffix as on overhang were designed to allow for the replacement of the standard BioBrick prefix and suffix with the Freiburg prefix and suffix.

Primers used:

Forward primer: 5'-GAATTCGCGGCCGCTTCTAGATGGCCGGCgcacaccatgaagaacaacac-3'

Reverse primer: 5'-CTGCAGCGGCCGCTACTAGTATTAACCGGTctcgtgatgcccgtggcag-3'

Capitals: Frieberg prefix or suffix lower-case: initial portion annealing to ends of NiBP.

This part is based on the existing BioBrick BBa_K1151001 by the 2013 Salente Lecce iGEM team. PCR was used to generate the part sequence as described above.
