The function of this part was characterised and quantified in a new salt-responsive device. The GFP encoded by <partinfo>BBa_E0040 </partinfo> was hooked up to an OmpR-responsive promoter (<partinfo>BBa_R0083</partinfo>) via a ribosome binding site (<partinfo>BBa_B0034</partinfo>) to produce a new composite part (a reporter) (<partinfo>BBa_K1012005</partinfo>). GFP fluorescence could be detected in an <i>E. coli</i> chassis, and we used an <i>envZ</i> mutant strain to show the sensor was working correctly.

We made an exact twin of this PqsR-encoding part and rather than send it to The Registry we decided to call our version <partinfo>BBa_K1157001</partinfo> and use it for our 2014 project. We added an HA epitope tag and showed that the protein was produced in an <i>E. coli</i> chassis (Dundee Figure 1). This part was used in conjunction with a <i>pqsA</i> promoter that we submitted as <partinfo>BBa_K1315001</partinfo>.

iGEM Dundee 2013

We made an exact twin of this PqsR-encoding part and rather than send it to The Registry we decided to call our version BBa_K1157001 and use it for our 2014 project. We added an HA epitope tag and showed that the protein was produced in an E. coli chassis (Dundee Figure 1). This part was used in conjunction with a pqsA promoter that we submitted as BBa_K1315001.
