MazF and LacI expression construct

MazF is a non-specific ribonuclease coding gene is used as killing agent against Bacillus subtilis. In our system, MazF is produced constitutively under Pveg promoter.

When MazF construct (Pveg + RBS SpoVG + MazF + RBS SpoVG + LacI + DT) was assemled in B.subtilis integration vector, BBa_K823023, we saw colonies when it is transformed into E.coli with Ampicillin resistance. On the other hand, we did not see any colony when the same plasmid was transformed into B.subtilis. Thus, it can be said that MazF construct is working in B.subtilis functionally.
