At the beginning Ptet promoter is on and HrpS forming rate is max,.So HrpS is adequate at that time. Once treated with protein TetR,concentration of HrpS is dropped to a low level. While there will not be HrpR unless treated with IPTG. Only when both HrpS and HrpR are at a high level, can the promoter hrpL trigger the transcription of following genes. This device function like a digital 'AND' gate.
