cI protein, which is a transcription factor,is a part of bandpass filter. It can repress the hybrid promoter K1031021[https://parts.igem.org/Part:BBa_K1031021].In order to fine-tune the band-pass filter,we construct a libiary of rbs before cI protein.

Component and Characterization

K1031013 consists of NahR[1], pSal promoter and transcription factor CƖ. CƖ functions as a repressor of hybrid promoter[2]. It is the repression branch of the Band-pass Filter circuit (Fig 1).

Fig 1 Repression circuit of Band-pass Filter. The orange square represents transcription factor NahR activating pSal promoter shown in light blue. Once pSal promoter is initiated, CƖ repressor, which is shown as square in dark blue, is expressed, inhibit hybrid promoter marked with two colors, thus inhibit expression of sfGFP.

Data shown

Previous work done by Dr. Lou Chunbo showed CƖ had obvious inhibiting effect to downstream promoter. (Fig 2).
