Hemeoxygenase with RBS
Synechocystis hemeoxygenase with a Ribosome Binding Site (RBS), oxidises Heme into biliverdin which absorbs light in the 360 nm and 650 nm wavelength producing a green colouration.
Usage and Biology
The BioBrick part in team Macquarie University (2011) mechanism
When coupled to a promoter BioBrick such as T7 promoter (BBa_I719005 and BBa_I712074) to produce parts BBa_K646003 and BBa_K646004, and transformed into Bl21(DE3)Tuner E. coli cells, a green colouration (require induction with ALA and IPTG) is produced.
Liquid media containing IPTG and ALA, incubated for 24 hours at low temperature, spun down. Pellets are green due to the production of biliverdin from the breakdown of heme - this indicates that heme oxyganse was expressed using T7 promoter.
