Long reporter bottom strand MammoBlock
Long reporter bottom strand to bind to Part:BBa_K779501 and Part:BBa_K779500.
Modifications: 3' ROX florophore, all bases modified with 2'O Methyl (ordered as a DNA oligo with Us instead of Ts).
IDT DNA Oligo Order: mUmGmGmAmUmAmGmUmUmGmGmUmGmGmGmAmGmAmGmGmAmGmUmGmGmGmUmG/3ROX_N/
This is an improved version of Part:BBa_K779116, with this design addressing the low melting temperature, orthogonality and small length of the previous iteration. Refer to Part:BBa_K779116 for data on the ROX fluorophore.
Usage and Biology
Provides a quenchable fluorophore. When annealed to a top strand (see parts above), the ROX fluorophore is quenched by an Iowa Black RQ quencher. When an input strand (Part:BBa_K779503) is added, strand displacement occurs and the quencher top strand is released.
