Enhanced tRNA Promoter for ''E. coli''
Usage and Biology
This tRNA terminator seqeuence is based around the downstream sequence of the E. coli Ala(GCC) tRNA. It contains the region of 3-6 T’s that is characteristic of tRNA terminators and an additional 10 bp of native sequence. This additional region of native sequence is to ensure RNA termination and to act as a small spacer region between tRNA genes.
It is important to use a tRNA terminator when expressing tRNA parts, as rho-dependent and rho-independent promoters rely on additional RNA sequence after the tRNA gene to function, and the addition of more RNA bases on a tRNA can severely affect its ability to function.
