T7Prom+HisTag+VioDC

This part contains a T7 promoter, lac operator, 6x his tag and the coding sequence for VioD and VioC (enzymes in the violacein production pathway).


Usage and Biology

A device was constructed that contained the two enzymes VioD and VioC which on their own do not produce a pigment. This is because the starting product of the reaction carried out by VioD and VioC is protodeoxyviolaceinic acid and is only made in the presence of VioA, VioB, and VioC.

However, when cells containing this construct were grown together with strains containing our VioABE construct BBa_K726016 , a purple pigment (violacein) was produced. More information on this process can be found on the 2012 UCSF iGEM Violacein Results Page .
