We tested the lac promoter through the Golden Gate Assembly Method. This promoter regulates the transcription of the lac operon in E.coli. We measured this promoter's strength as a measure of fluorescence under the 7 different following conditions: a positive (plasmid, red fluorescent protein, and RBS) and two negative controls (broth and plasmid without promoter), a small culture with and without IPTG, and a large culture with and without IPTG. IPTG is a compound that triggers the transcription of the lac operon.

Applications of BBa_J100094
