CRP protein binds cAMP, which causes a conformational change that allows CRP to bind tightly to a specific DNA site in the promoters of the genes it controls. Often, CRP is considered as an activator. In our design, we successfully turn the activator into repressor by altering the position of binding site in the CRP regulated promoter. This device contains a constitutive promoter J23116 .
